چکیده

A simplified procedure for the identification and measurement of single-ring aromatic products of lignin acidolysis is described. The procedure employed a 6-h hydrolysis of spruce milled wood lignin in acidic dioxane at 87 degrees C, followed by a series of organic extractions to recover acidolysis products which were quantified by gas chromatography of trimethylsilyl derivatives. Complex gel permeation chromatography procedures utilized by other workers were avoided in the modified procedure, but equivalent results were obtained. The simplified procedure was utilized to hydrolyze sound and actinomycete-decayed spruce milled wood lignins and was shown to be useful as a technique for the rapid screening of microorganisms for their ability to alter lignin.

Determination of lignin-modifying enzymes (LMEs) in Hyphodermella species using biochemical and molecular techniques

White-rot basidiomycetes are one of the most important lignolytic microorganisms. These fungi have been reported to secrete three main classes of lignin degrading enzymes: lignin peroxidases (LiPs), manganese peroxidases (MnPs) and laccases. In this study, for the first time the lignin degrading capability of two plant pathogens i.e. Hyphodermella rosae and H. corrugata was evaluated using both...

Lignin-solubilizing ability of actinomycetes isolated from termite (Termitidae) gut.

The lignocellulose-degrading abilities of 11 novel actinomycete strains isolated from termite gut were determined and compared with that of the well-characterized actinomycete, Streptomyces viridosporus T7A. Lignocellulose bioconversion was followed by (i) monitoring the degradation of [14C]lignin- and [14C]cellulose-labeled phloem of Abies concolor to 14CO2 and 14C-labeled water-soluble produc...

The Discovery and Promise of Lignin-degrading Enzymes

This paper briefly summarizes our research at the U.S. Forest Products Laboratory aimed at understanding how lignin is degraded by microorganisms. Our ultimate purpose is to gain the background knowledge needed to use lignin-degrading microbes and their enzymes in wood processing. Today such research would fall within the realm of biotechnology, although when we started that word had not yet be...
